NY 110 begins at Merrick Road (NY 27A) in Amityville. The first section is known as Broadway and is a two-lane undivided road.

North of the parkway, NY 110 has six lanes and is called Broad Hollow Road. There is a traffic light at CR 47. Turn left onto Main Street to go north. Turn right onto Great Neck Road to go south. This time, there actually is county route signage.

Broad Hollow Road continues north from Farmingdale to Melville. It passes under the Main Line of the Long Island Rail Road. There are traffic lights at Melville Road, Milbar Boulevard, Walt Whitman Road, and Duryea Road. Ruland Road (CR 5) also begins on the right at one of the traffic lights, but there is no county route signage for it. There are a number of reassurance shields along this stretch, including another erroneous US 110 shield.

There is a traffic light at Pinelawn Road (CR 3). Turn left to go north or right to go south. Sweet Hollow Road begins one block to the left at the northern terminus of Pinelawn Road (CR 3). The signage here is a little misleading because it makes it look like Sweet Hollow Road begins at this traffic light.
