NY 25 is a main east/west route for northern Long Island. The first section of NY 25 is the Queensboro Bridge, a bridge over the East River from Manhattan to Queens. It has two roadways. On the Queens side of the bridge, NY 25 runs to the southeast as Queens Boulevard, a large divided road through the western half of Queens. (The lower roadway of the bridge leads directly to Queens Boulevard. The upper roadway feeds onto Thomson Avenue, which joins Queens Boulevard slightly further to the east.)

NY 25 leaves Queens Boulevard at Hillside Avenue and runs to the east along Hillside Avenue. NY 25 leaves Hillside Avenue at Braddock Avenue and is routed along Braddock Avenue. (Hillside Avenue east of this point is designated NY 25B.) Braddock Avenue runs to the southeast until the Queens-Nassau county line, where it ends at Jericho Turnpike. Jericho Turnpike runs to east through Nassau and western Suffolk. In Smithtown, NY 25 and NY 25A are multiplexed as Main Street. When they split, NY 25 is called Middle Country Road. It keeps this name until Riverhead. After Riverhead, NY 25 is called Main Road and it runs along the North Fork as a small two-lane road. It ends at the ferry terminal at Orient Point, the easternmost point on the North Fork.
