US 50 enters Virginia multiplexed with I-66 across a bridge over the Potomac River. On the other side of the bridge, the two routes split.
The first section of US 50 in Virginia is a large divided highway with service roads called Arlington Boulevard. It has a mixture of grade-separated interchanges and traffic lights. There is an exit for Rosslyn and the Key Bridge (US 29).

Skipping many miles to the west, there is a section just west of Middleburg where John Mosby Highway widens to become a four-lane divided road. After a short distance, the median ends and it narrows back down to two lanes.

John Mosby Highway widens to become a four-lane divided road again. There is a traffic light at US 17. Turn left onto Winchester Road to go south. US 17 north is multiplexed with US 50 west along John Mosby Highway straight ahead.

After a short distance, there is a traffic light at Lord Fairfax Highway (US 340). Turn right to go north or left to go south.

There is a pair of traffic lights at I-81 and US 522. Turn left onto Front Royal Pike at the first light to go south on US 522. Turn right at the first light to go north on I-81 or left at the second light to go south. Northbound US 522 is multiplexed with US 17 and US 50 straight ahead.

Skipping a few blocks, Millwood Avenue runs north through Winchester as a two-lane undivided road. There is a railroad crossing.

There is a traffic light at South Cameron Street (US 11). Turn right go north and to stay on US 17/US 50/US 522, which is multiplexed with US 11 beginning here. To go south on US 11, continue straight ahead onto Gerrard Street.
