Baltimore Pike runs northeast through Glen Mills. There are traffic lights at Concord Road, Cheyney Road, Powell Road, Stoney Bank Road, Lydia Hollow Drive, Walnut Hill Boulevard, Valleybrook Road, and Red Roof Drive.

There is a bridge over Chester Creek. The roadway is temporarily narrowed down to one lane in each direction due to construction.
