NY 25B begins at Jericho Turnpike (NY 25) and runs west as a four-lane divided road.

There is a traffic light at Glen Cove Road (CR 1). Turn right to go north or left to go south.

Hillside Avenue passes over the Northern Parkway, but there is no entrance or exit. It narrows down to two lanes and the divider ends.

The two-lane section in East Williston is called East Williston Avenue. It is locally maintained by the Nassau County Department of Public Works (NCDPW) and is designated NY 25B/CR 85.

There is a traffic light at Roslyn Road (CR 7A). Turn right to go north or left to go south.

East Williston Avenue continues east for a short distance further. There is a railroad crossing at the Oyster Bay Branch of the Long Island Rail Road. CR 85 ends at the railroad crossing, and East Williston Avenue becomes Hillside Avenue again.
