Riverhead-Hampton Bays Road begins at Montauk Highway (CR 80) as a four-lane divided road and runs north. There is a cloverleaf interchange with Sunrise Highway (NY 27).

The divider ends, and Riverhead-Hampton Bays Road narrows down to two lanes and curves to the northwest. The Big Duck is on the right along this section.
