MA 3 is a north/south highway connecting Cape Cod to the Boston area. It begins on the west side of the Sagamore Bridge (US 6). The bridge roadway becomes MA 3, as US 6 takes an exit onto Scenic Highway. The first section of MA 3 is an expressway called Pilgrims Highway. It is so named because it passes by Plymouth, where the Pilgrims famously arrived in Massachusetts from England aboard the Mayflower, as celebrated on the American holiday of Thanksgiving.

Pilgrims Highway ends at the Southeast Expressway (I-93) in Braintree. From there, I-93 and MA 3 are multiplexed north to Boston, where they splits. MA 3 follows Embankment Road, a short highway along the Charles River in Boston. MA 28 joins it after one exit. After a short distance, MA 3 splits off. Embankment Road continues west for one more exit as MA 28 before becoming Storrow Drive.

The final section of MA 3 is the Longfellow Bridge, which crosses the Charles River to Cambridge. On the other side, MA 3 follows Memorial Drive for a short distance to its interchange with Massachusetts Avenue (MA 2A). There, the designation changes to US 3.
