CR 526 begins at Hightstown Road (CR 571) in West Windsor. While it is designated an east/west route, its first section runs south for a few miles. It is initially called South Mill Road and changes names to become New Edinburg Road and then Edinburg Road. It turns west onto Old Trenton Road (CR 535) for one block and then south again onto Robbinsville-Edinburg Road, which extends for a couple of miles to end at NJ 33. The two routes are multiplexed for one block, and then CR 526 splits off to the east onto Robbinsville-Allentown Road.
After a few blocks, there is a traffic light at US 130. CR 526 turns north onto US 130 and then splits off at the next traffic light onto the Robbinsville Bypass, which runs north of the downtown area of Robbinsville and then curves to merge with Robbinsville-Allentown Road. CR 526 follows Robbinsville-Allentown Road, which crosses the New Jersey Turnpike (I-95) with no interchange and then curves to the south to pass through an interchange with I-195.
In Allentown, Robbinsville-Allentown Road becomes Church Street, which ends at Old York Road (CR 524). After one block, it splits off onto Waker Avenue. Heading out of town, Waker Avenue becomes Trenton-Lakewood Road. In Millstone, CR 571 joins CR 526 along Trenton-Lakewood Road. Upon crossing Monmouth Road (CR 537), it changes names to become West Commodore Boulevard. After a short distance, the two routes split. CR 526 turns east to stay on West Commodore Boulevard, while CR 571 continues south as Cassville Road.
At Cedar Swamp Road (CR 527), West Commodore Boulevard becomes East Commodore Boulevard, which ends after a short distance at Jackson Mills Road (CR 638). The two routes are multiplexed for one block, and then CR 526 splits off onto County Line Road, which runs southeast to Lakewood. At Ridge Avenue (CR 623), it changes names to become Lanes Mill Road. It ends at CR 549, which, rather confusingly, is also called Lanes Mill Road in the area.
