MA 129 is an east/west route through the suburbs north of Boston. It begins in Chelmsford at the intersection where MA 110 turns from Littleton Road onto Chelmsford Street. The first section is known as Billerica Road. It runs southeast to the US 3 expressway and changes names to become Chelmsford Road. After a short distance, Chelmsford Road ends at Boston Road (MA 3A). The two routes are multiplexed for a short distance to the south, and then MA 129 splits off to the east onto Floyd Street, which changes names to become Salem Road. As it continues east, it changes names again to become Shawsheen Avenue and then Richmond Street, ending at Main Street (MA 38). Once again, the two routes are multiplexed to the south, and then MA 129 splits off onto Lowell Street. After about a mile, there is an interchange with I-93. After another mile, it crosses Main Street (MA 28) in Reading and changes names to become Salem Street.

After a short distance, there is an interchange with I-95. On the other side, MA 129 is known as Lowell Street again. After a couple of blocks, it turns south onto Main Street, which runs south along the east side of Lake Quannapowitt into the downtown area of Wakefield. There, it turns east again onto Water Street. After a couple of miles, Water Street ends at Walnut Street. MA 129 turns south onto Walnut Street and follows it for a short distance to US 1.

MA 129 follows US 1 north for one exit and then splits off onto Salem Street. After about half a mile, there is a rotary with access to I-95. MA 129 leaves the rotary on Lynnfield Street, which runs southeast to Lynn and changes names to become Broadway. MA 129 turns south onto Boston Street and then east onto Washington Street, which ends at Broad Street. MA 129 turns east onto Broad Street, which changes names to become Lewis Street, then Humphrey Street, then Atlantic Avenue as it makes it way east to Marblehead. Then, it turn north onto Ocean Avenue for two blocks to end at Pleasant Street (MA 114). The two routes end at east other, although Pleasant Street continues east as a local street.
