NY 49 is an east/west route extending from Fulton, which is located along the Oswego River northwest of Syracuse, to Utica. It begins at NY 3 and runs east to Oneida Lake and then along the north side of the lake. A few miles east of the lake, NY 49 joins NY 46. The two routes are multiplexed along Rome-New London Road, Erie Boulevard West, and Erie Boulevard East to Rome, where they split. For the rest of its length, NY 49 is an expressway called the Utica-Rome Expressway, which follows the Erie Canal southeast to Utica. There, it merges with I-790/NY 5, which becomes the service road of the New York Thruway (I-90).
